RESUMO

The hydrothermal method was used to create dilute magnetic semiconductor nanoparticles of Zn1-x Co x O (x = 0, 0.01, 0.05, 0.09). The effect of cobalt doping on the microstructure, morphological and optical properties of Zn1-x Co x O was also studied and the Co doping to host ZnO was confirmed from XRD and EDX analysis. The structural analysis showed that doping of cobalt into ZnO decreased the crystallinity, but the preferred orientation didn't change. SEM analysis revealed that the cobalt dopant did not have a strong influence on the shape of the synthesized nanoparticles. No defect-related absorption peaks were observed in the UV-Vis spectra. The crystallinity of the doped samples was improved by high growth temperature and long growth time. Ferromagnetic behavior above room temperature was detected in co-doped ZnO nanoparticles. The ferromagnetic behavior increased with increasing Co (up to x = 0.05) doping. The ferromagnetic behavior declined when the Co content was further increased. Related research shows that doped ZnO nanoparticles have better dielectric, electrical conductivity, and magnetic properties than pure ZnO. This high ferromagnetism is usually a response reported for dilute magnetic semiconductors. These semiconductor nanoparticles were further used to designed spintronic based applications.

BACKGROUND AND PURPOSE: Although drowning is a leading cause of mortality and morbidity in young children, the neuropathologic consequences have not been fully determined. The purpose of this article was to quantitatively characterize white matter microstructural abnormalities in pediatric anoxic brain injury from nonfatal drowning and investigate the correlation with motor function. MATERIALS AND METHODS: Whole-brain T1-weighted and diffusion-weighted MR imaging datasets were acquired in 11 children with chronic anoxic brain injury and 11 age- and sex-matched neurotypical controls (4-12 years of age). A systematic evaluation form and scoring system were created to assess motor function. Tract-Based Spatial Statistics was used to quantify between-group alterations in the diffusion tensor imaging indices of fractional anisotropy and mean diffusivity and to correlate with per-subject functional motor scores. RESULTS: Group-wise Tract-Based Spatial Statistics analyses demonstrated reduced fractional anisotropy in the bilateral posterior limbs of the internal capsule and the splenium of the corpus callosum (P < .001). Mean diffusivity was more diffusely increased, affecting the bilateral superior corona radiata, anterior and posterior limbs of the internal capsule, and external capsules (P < .001). Individual-subject fractional anisotropy and mean diffusivity values derived from the ROIs of the bilateral posterior limbs of the internal capsule strongly correlated with motor scores and demonstrated more potent between-group effects than with ROIs of the entire corticospinal tract. CONCLUSIONS: These data particularly implicate the deep white matter, predominantly the posterior limbs of the internal capsule, as targets of damage in pediatric anoxic brain injury with drowning. The substantial involvement of motor-system tracts with relative sparing elsewhere is notable. These results localize white matter pathology and inform future diagnostic and prognostic markers.

BACKGROUND AND OBJECTIVE: Interleukin (IL) 4 is a cytokine that mediates allergic responses. Different single nucleotide polymorphisms (SNPs) can influence the immune response mediated by cytokines. The aim of the present study was to investigate the possible association between IL-4 polymorphisms and allergic rhinitis and atopic asthma. METHODS: A total of 214 atopic patients (108 with asthma and 106 with allergic rhinitis) and 120 healthy controls from Pakistan were genotyped for IL-4 SNPs C-589T (rs2243250), T+2979G (rs2227284), and C-33T (rs2070874) using restriction fragment length polymorphism-polymerase chain reaction. Statistical analysis was performed using the statistical software package StatCalc, Epilnfo v.6. RESULTS: The SNP rs2243250 was significantly associated with both asthma (P = .004, chi2 = 11.0) and allergic rhinitis (P < .001, chi2 = 20.2), as was T-2979G (P < .001, chi2 = 22.51 for asthma and P < .001, chi2 = 57.6 for allergic rhinitis). The most frequent genotypes in the asthma and allergic rhinitis groups were TT for SNP rs2243250, and GG for SNP rs2227284. rs2070874 was not found to be associated with either of the 2 atopic respiratory diseases analyzed in the Pakistani cohort. CONCLUSIONS: rs2243250 and rs2227284 are significantly associated with asthma and allergic rhinitis. The results of this study indicate that in addition to environmental factors, genetic risk factors also play an important role in the development of atopic respiratory diseases.

Introducción y objetivo: La IL-4 es una citocina que media las reacciones alérgicas. Diferentes polimorfismos en nucleótidos simples (SNPs) pueden influir sobre la respuesta mediada por citocinas. El motivo de este trabajo fue investigar la posible asociación de polimorfismos de la IL-4 con rinitis alérgica (RA) o asma atópica. Método: Se incluyó un total de 214 pacientes atópicos (asma n=108, RA n=106) y 120 controles sanos de Paquistán que fueron genotipados para SNPs IL-4 C-589T (rs2243250), T+2979G (rs2227284) y C-33T (rs2070874) mediante PCR. El análisis estadístico se llevó a cabo mediante el paquete StatCalc, EpiInfo v.6. Resultados: Observamos que el SNPs rs2243250 se asocia de forma significativa a asma (p 0.004; X2=11.0) y a RA (p<0.001; X2=20.2). Los genotipos más frecuentes en asma y RA fueron TT para SNP rs2243250, y GG para SNP rs2227284. Por otra parte, el SNP rs2070874 no se asocia con ninguna enfermedad respiratoria en una cohorte pakistaní. Conclusiones: rs2243250 y rs2227284 se asocian significativamente a asma y a RA. Los resultados de este estudio demuestran que además de los factores ambientales, los factores genéticos de riesgo juegan un papel importante en el desarrollo de las enfermedades atópicas respiratorias (AU)

Liposome co-encapsulation of synergistic anti-cancer drug combination is an emerging area that has demonstrated therapeutic benefit in clinical trials. Remote loading of two or more drugs into a single liposome constitutes a new challenge that calls for a re-examination of drug loading strategies to allow the loading of the drug combination efficiently and with high drug content. In this study, the Mn(2+) gradient coupled with A23187 ionophore was applied in the sequential co-encapsulation of doxorubicin and irinotecan, as this drug loading method is capable of remotely loading drugs by apparently two different mechanisms, namely, coordination complexation and pH gradient. Doxorubicin and irinotecan could be co-encapsulated into liposomes in a wide range of drug-to-drug ratios, with encapsulation efficiencies of > 80%. The total encapsulated drug content was non-linearly correlated with increases in the intraliposomal Mn(2+) concentration, with a maximum total drug-to-lipid molar ratio of 0.8:1 achieved with 600 mM Mn(2+). This high encapsulated drug content did not affect the stability of the co-encapsulated liposomes upon storage for six months. Regardless of the encapsulated drug amount, the liposomes did not exhibit the fiber bundle precipitate morphology but rather an undefined structural organization in the aqueous core. The co-encapsulated liposome formulation was further tested in an intraperitoneally grown, human ovarian tumor xenograft model, and was shown to significantly improve the survival of the tumor-bearing animals. The improvement in therapeutic efficacy was possibly due to the increase in systemic drug exposure, with the maintenance of the synergistic molar drug ratio of 1:1 in circulation.

BACKGROUND: Several studies have shown that promoter polymorphisms of the CD14 gene are associated with atopic asthma. However, the results of association studies in different populations are conflicting. This study aimed to investigate the possible association between the CD14 polymorphisms A-1145G and C-159T and atopic phenotypes in Pakistani cohorts. METHODS: Healthy controls (n = 120) and atopic patients (n=220) were genotyped for the single-nucleotide polymorphisms C-159T (rs2569190) and A-1145G (rs2569191) using restriction fragment length polymorphism-polymerase chain reaction. RESULTS: The genotype and allelic frequencies were in Hardy-Weinberg equilibrium. Overall, strong associations were observed between both C-159T (P = .02; chi2 = 7.16) and A-1145G (P = .01; chi = 7.88) and atopy. The G allele of A-1145G was significantly associated with atopy (P < .009; chi2 = 6.72). When the data were stratified, the associations observed were due to the individual phenotypes: atopic asthma was significantly associated with A-1145G (P = .02; chi2 = 7.18), whereas the association between C-159T and atopy was attributed to patients with allergic rhinitis (P = .01; chi2 = 8.13). CONCLUSION: In Pakistani adults, the A-1145G polymorphism is associated with atopic asthma, whereas the C-159T polymorphism is significantly associated with allergic rhinitis.

The use of drug cocktails has become a widely adopted strategy in clinical cancer therapy. Cytotoxic drug cocktails are often administered based on maximum tolerated dose (MTD) of each agent, with the belief of achieving maximum cell kill through tolerable toxicity level. Yet, MTD administration may not have fully captured the therapeutic synergism that exists among the individual agents in the drug cocktail, as the response to a cocktail regimen, that is, whether the effect is synergistic or not, could be highly sensitive to the concentration ratios of the individual drugs at the site of action. It is important to realize that the inherently different pharmacokinetic profiles of the individual agents could have significant influence on the response to an anti-cancer drug cocktail by dictating the amount of the individual agents reaching the tumor site and therefore the concentration ratios. Furthermore, the individual agents may have unfavorable pharmacokinetic interactions that add to the difficulty in determining the therapeutic and/or toxicological effects of the drug cocktail. In this review, we will focus on how lipid-based nanoparticulate systems could address the above issues associated with anti-cancer drug cocktails. Specifically, we will highlight the use of liposome systems as the means to control and coordinate the delivery of various anti-cancer drug cocktails, encompassing conventional chemotherapeutics, chemosensitizing agents and molecularly targeted agents.

PURPOSE: In obstetric brachial plexus lesions, muscle imbalance caused by active supinator muscles and paralyzed pronator muscles can result in a supination position of the wrist, which, apart from cosmesis, may interfere with function. METHODS: In this retrospective study, we describe the results of a pronating radius osteotomy for supination deformity of the hand in children with an obstetric brachial plexus lesion. RESULTS: After a mean follow-up of 23 months, all 8 patients (mean age, 9.4 years; range, 4-13 years), operated between 1998 and 2006, had improved functionally and aesthetically. CONCLUSIONS: All patients had improved functionally and aesthetically.

We report a case of L5 radiculopathy secondary to intrapelvic placement of an acetabular screw impinging on the lumbosacral trunk. A 43-year-old man underwent revision of an uncemented total hip replacement to a custom-made femoral component and uncemented cup. He subsequently developed hip pain, dysesthesia, and L5 radiculopathy. Spiral computed tomography (CT) and plain radiographs raised the possibility of an acetabular screw abutting the neurovascular bundle within the pelvis. Operative removal of the intrapelvic portion of the screw resulted in the immediate relief of the patient's radicular symptoms. This case report highlights the importance of the positioning of intrapelvic screws and the need for an anatomic awareness of the structures at risk during acetabular screw placement.

To describe a new method of catheter insertion in intradiscal electrothermal therapy, which eliminates the need for reinsertion of the cannula and catheter from the contralateral side in those patients in whom optimal positioning is not achieved with the standard technique. This new technique has not been described before. In those patients in whom adequate catheter position cannot be achieved with the standard technique, instead of withdrawing the cannula after the initial treatment, we recommend rotating the cannula 180 degrees through its long axis. This will allow the catheter to hit the anterior anulus and deflect backwards toward the cannula. It can then be negotiated across the midline to adequately thermally treat the whole posterior anulus. We have performed our technique in 42 consecutive patients in whom initial navigation was difficult. This new method proved to be simple and did not cause patients additional discomfort. The "pig tail" technique is safe and effective in intradiscal electrothermal therapy of those patients with difficult navigation. It avoids the need for second needle insertion, therefore avoiding the use of more local anesthesia, further discomfort for the patient, and additional radiographic exposure.

The anatomical parameters of the thoracic and lumbar regions of the deer spine were evaluated and compared with the existing data of the human spine. The objective was to create a database for the anatomical parameters of the deer spine, with a view to establish deer spine as a valid model for human spine biomechanical experiments in vitro. To date, the literature has supported the use of both calf and sheep spines as a suitable model for human spine experiments as the difficulty in procuring the human cadaveric spines is well appreciated. With the advent of Bovine Spongiform Encephalopathy (BSE) and its likely transmission to human in form of new variant Creutzfeld Jakob disease (CJD), there is a slight risk of transmission to humans through food chain if proper precautions for disposal of specimen are not adhered to. There is also a significant risk of transmission through direct inoculation to the researchers (Wells et al. Vet. Rec., 1998:142:103-106), working with infected bovine and sheep spine. The deer spines are readily available and there are no reported cases of deer being carriers of prion diseases (Ministry of Agriculture, Fisheries and Food, 1998). Six complete deer spines were measured to determine 22 dimensions from the vertebral bodies, endplates, disc, pedicles, spinal canal, transverse and spinous processes, articular facets. This was compared with the existing data of the human spine in the literature. The deer and human vertebrae show many similarities in the lower thoracic and upper lumbar spine, although they show substantial differences in certain dimensions. The cervical spine was markedly different in comparison. The deer spine may represent a suitable model for human experiments related to gross anatomy of the thoracic and lumbar spine. A thorough database has been provided for deciding the validity of deer spine as a model for the human spine biomechanical in vitro experiments.

Mycobacteria generally have thick cell walls and contain large amounts of lipid, making them resistant to DNA extraction. Five methods, namely, extensive enzymic digestion method (M1), 2-min mechanical glass-bead disruption method (M2), thermal shock method (M3), modified conventional enzymic digestion method (M4), and manual disruption with modified conventional enzymic digestion method (M5), were used to compare their effectiveness and simplicity in extracting DNA from slowly growing mycobacteria (Mycobacterium leprae, M. lepraemurium and M. bovis BCG), and a rapidly growing mycobacterium (M. phlei). The highest DNA yield was obtained by M2 from M. lepraemurium which produced 2.82 micrograms DNA/mg wet weight of cells, representing a theoretical yield of 78%. M3 gave the lowest DNA yield; 0.01 microgram DNA/mg wet weight of cells of M. lepraemurium was obtained. M4, in which proteinase K was used, is more effective than M1, in which subtilisin and pronase were used. M5 yielded a higher amount of DNA, but it required more manipulations to extract DNA as compared to M4. Extraction of DNA of M. leprae from nude mice is more difficult than that of M. leprae from armadillos by all of the methods used. These results suggest that the biosynthetic capabilities of these two forms of M. leprae may vary, depending on their cultural conditions and/or strain differences. Our results have shown that both M2 and M4 are the simplest, most effective and time-saving methods which are suitable for every routine laboratory to extract DNA from slowly and rapidly growing mycobacteria.

RESUMO

The susceptibility of severe combined immunodeficient (SCID) mice to human leprosy bacilli was investigated. Nude mice were used as controls. SCID mice were found to be highly susceptible to Mycobacterium leprae and the progress of infection was comparatively 2-3 months earlier than observed in the nude mice. After reaching a maximum of approximately 1 x 10(9) acid fast bacilli/foot pad at about 8 months postinfection the number of bacilli gradually decreased. The progress of infection in nude mice was different from that found in SCID mice. The multiplication of M. leprae in the foot pads of nude mice continued and reached approximately 2.0 x 10(10) bacilli/foot pad and then nearly remained the same. The results indicate that SCID mice can be used as a suitable model for screening antileprosy drugs while nude mice should be involved in the production of M. leprae for use in other fields of leprosy research.

RESUMO

Trauma in elderly patients is often difficult to evaluate and neck injuries are probably more common than is documented. A case is described of a missed odontoid peg fracture with the unusual presentation of dysphagia and a rapidly developing cervicodorsal kyphos (head falling forward), which may well have been anticipated at several points in the management chain. The lessons to be learned are outlined, together with the subsequent management of this patient.
